【摘要】 目的 比较清热合剂各提取部位群的解热抗炎药效作用。方法 分别采用2,4二硝基苯酚致热大鼠模型、内毒素致热家兔模型、二甲苯致小鼠耳肿胀及蛋清致大鼠足肿胀模型,研究清热合剂各提取部位群的解热、抗炎作用。结果 清热合剂的20%、40%和60%乙醇提取部位群均有解热抗炎作用,其中清热合剂的40%乙醇提取部位群解热抗炎作用最强。结论 清热合剂的40%乙醇提取部位群具有明显解热抗炎作用,作用效果与清热合剂原液相当。

Abstract:Objective To investigate the feverrelieving and antiinflammatory effects of Qingre mixture (QR) for oral use.Methods Animal models of ear edema induced by xylene in mice,paw edema induced by egg white and fever induced by 2,4dinitrophenol in rats and endotoxininduced fever in rabbits were made; the feverrelieving and antiinflammatory effects of the different extractions of Qingre mixture were studied.Results Extractions of Qingre mixture extracted by 20%,40%,and 60% ethanol showed a marked suppressive effects on ear edema induced by xylene in mice,paw edema induced by egg white and fever induced by 2,4dinitrophenol in rats and endotoxininduced fever in rabbits,the extraction of Qingre mixture extracted by 40% ethanol was the most effective.Conclusion Extraction of Qingre mixture extracted by 40% ethanol had feverrelieving and antiinflammatory effects,which were equivalent to the effects of Qingre mixture.

清热合剂是由黄芩、葛根、大青叶等五味中药组成的传统中药复方制剂,具有疏散风热、清热解毒、利咽止咳的功能,用于病毒性感冒或流行性感冒所见身热、头胀痛、咽喉红肿疼痛、鼻塞等症的治疗[1,2]。但由于合剂携带服用不便,久储有较多沉淀,外观性状较差和难以作为新药进入药品市场等缺点,本研究拟通过大孔吸附树脂对清热合剂粗提物进行分离纯化后将其研制成颗粒剂,使之药效更明确,降低不良反应的发生率。为了明确清热合剂各提取部位群的药效变化,确保其质量和疗效,本文通过建立大鼠、家兔发热模型、二甲苯致小鼠耳肿胀及蛋清致大鼠足肿胀模型,比较原清热合剂及分离纯化后各提取部位群的解热及抗炎作用,为确定清热合剂的有效部位群提供依据。
1 材料
1.1 药品与试剂
清热合剂(QR,ρ=1.35 g·mL-1),中国医科大学附属盛京医院药剂科生产,批号:20071207;阿司匹林肠溶片(50 mg/片),沈阳万隆药业有限责任公司,批号:071002;醋酸地塞米松片(5 mg/片),天津药业基团有限公司,批号:070325;二甲苯,天津市科盟化工工贸有限公司,批号:050480;内毒素,规格500 ng/2500 Eu/支,上海申索试剂有限公司,批号:20070417;羧甲基纤维素钠(CMCNa),上海昌为医药辅料技术有限公司。
1.2 仪器
HHS电热恒温水浴锅:江苏红旗医疗器械厂;MC107BW型欧姆龙电子体温计:欧姆龙大连有限公司。
1.3 动物
昆明种小鼠,雌雄各半,体质量25~30 g;Wistar大鼠,雄性,体质量120~150 g、180~220 g;新西兰大耳白兔,雌雄各半,体质量1.8~2.2 kg。均由中国医科大学附属盛京医院医学实验动物中心提供,合格证号:辽实动质字SCXK[辽]2003013号。
